Description

(Magnetic Beads Method)

【Product Introduction】

The paraffin-embedded tissue DNA extraction reagent uses an environmentally friendly deparaffinization method to remove paraffin, and special lysis conditions are applied to release the DNA in the tissue section, overcoming the inhibition effect caused by formalin cross-linking. The reagent uses DNA-binding magnetic beads and a unique buffer system to purify high-quality DNA while maximizing the removal of proteins, inorganic salts and other organic impurities. The obtained DNA has good integrity, high purity, stable and reliable quality, and is suitable for downstream experiments such as PCR, qPCR, SNP gene analysis, and sequencing.

【Product advantages】

1. Safe and non-toxic: no need for xylene dewaxing, no need for toxic organic reagents such as phenol/chloroform

2. Excellent applicability: suitable for paraffin sections of various tissues

3. High purity: effectively remove PCR inhibitors such as proteins and inorganic salts, which is convenient for downstream experiments.

4. This kit can be used for manual extraction, and can also be integrated with a variety of high-throughput extractors.

【Experimental case】

Case1:

The DNA of 10 cases of paraffin-embedded tissues (5 μm, 6 sections) of human gastric mucosa was extracted by using our company’s paraffin-embedded tissue DNA extraction kit, and then qPCR was performed with human GAPDH housekeeper gene as the target gene, and whether the extracted product affected qPCR amplification.

Result analysis: The results showed that the amplification curve was S-shaped, and there was no obvious inhibition. The results show that the DNA extracted by this reagent is of excellent quality and can be directly used in downstream PCR and other follow-up experiments.

Case2

The DNA of paraffin-embedded tissue DNA extraction reagent was used to extract the paraffin-embedded tissue (20 μm, 1 sections) of pig lung, canine pancreas, and chicken lung, and the extracted products were tested by ultra-micro-ultraviolet-visible spectrophotometer, and the results were as follows:

No.SampleA260/A280A260/A230Conc.(ng/μL)Yield(μg)
1Porcine lung embedded tissue sections1.752.104297.129.71

5Chicken lung embedded tissue sections1.6161.946114.811.48

Results: The results showed that the reagent could stably extract DNA with good purity and high yield for different types of embedded tissues, which could be directly used in downstream PCR and other follow-up experiments.

【Application area】

The purified DNA extracted from this kit can be directly used in downstream molecular biology experiments such as PCR, qPCR, dPCR, NGS, SNP gene analysis, STR gene identification, pathological molecular research, etc., and can be applied to identification, disease diagnosis, personalized medicine and other fields.
