【Experimental cases】

Case: 

• The sample is DH 5α E. coli (containing PHY300PLK-egls gene plasmid, base pair about 6000 bp, low copy plasmid), bacterial solution OD₆₀₀ = 0.032. Plasmid was extracted by LJbio192H fully automated nucleic acid extraction and purification instrument. After extraction was determined by Quawell Q5000 trace ultraviolet visible spectrophotometer, the results are as follows:

Note: the original concentration of 6 μL on the sample electrophoresis, agarose gel concentration of 1%, 120 V electrophoresis.

(2) Results analysis:

Quawell Q5000 The results of microultraviolet visible spectrophotometer show that:

① The value of  A₂₆₀ / A₂₈₀ is 1.82-1.832 and A₂₆₀ / A₂₃₀ is 2.06-2.081, demonstrating the high purity of the extracted product.

② Under OD₆₀₀ = 0.032, the yield of 50 mL of culture solution can reach about 97.63 µg.

The results of the agarose gel electrophoresis then showed that:

The bands are bright and complete.

In conclusion, 50 mL of endofree DNA was extracted from plasmid, producing the products with high purity, high yield and complete electrophoretic bands.
