【Product Introduction】

This product is designed for the extraction, enrichment, and purification of genomic nucleic acids from fresh, frozen, or blood samples containing anticoagulants. Under the influence of guanidine salts, cellular lysis occurs, releasing the nucleic acids. Through specific adsorption by magnetic beads, the nucleic acids are separated from other contaminants in the sample. They are then washed with a buffer solution to eliminate proteins and residual guanidine salts. Ultimately, purified nucleic acids are retrieved via elution and magnetic adsorption. The resulting product is suitable for applications such as PCR amplification, nucleic acid detection assays, gene mutation analysis, and sequencing experiments.

Genomic DNA was extracted from fresh blood samples of healthy individuals using a Blood Genomic DNA Extraction Reagent kit on the automated nucleic acid extraction and purification instrument, LJbio32H. The electrophoresis results are shown in the following figure.

Sample volume:200 μL, Elution volume :80 μL, electrophoresis loading volume:6 μL,Agarose gel concentration is 1%,120 V,time:30 mins.

Result analysis:

From the extracted product detection data and agarose electrophoresis results, it can be seen that:

1. The extraction time is about 20 minutes, and all 14 samples can stably extract human whole blood genomic DNA;

2. The purity of the extracted products is good: the 260/280 ratios for all extraction products are above 1.9, and the 260/230 ratios are above 1.4.

3. High yield of extraction products: Yields from all 200 μL samples are consistently above 3 μg, sufficient to meet the requirements of downstream applications.

4. The electrophoretic bands are complete and without degradable and diffuse.

Result analysis:The test results of the micro nucleic acid analyzer indicate that:

  1. The coefficient of variation of A260/A280 and A260/A230 below 2%.The test results demonstrate high stability of the reagent in extracting nucleic acids.。
  2. The 260/280 ratios for all extraction products between 1.906 and 1.944,the 260/280 ratios for all extraction products between 1.892 and 2.04.The intact and nearly coinciding peak profiles attest to the high purity of the extracted product.。
  3. The extraction amount of nucleic acid from 200 µL pig whole blood can reach over 8.25 µ g.

The results of agarose gel electrophoresis showed that:

Seven samples were randomly selected, all exhibiting bright bands with good integrity.

In summary, 32 samples of whole blood were extracted using a fully automatic nucleic acid extraction and purification instrument. The extracted products had high purity, high yield, complete electrophoresis bands, and uniform and stable combination of reagents and instruments.
