Improve the Tissue Sample Pretreatment Process to Ensure the Quality of RNA Extraction

Tissue samples are an indispensable part of biomedical research, and extracting high-quality RNA from tissues is crucial to the success of many experiments. The following pretreatment considerations can help you maximize the quality of RNA in tissue samples.

I. Shorten the cold ischemia time

The cold ischemia time refers to the time interval between tissue removal and storage. The shorter the time is, the lower the risk of RNA degradation. Therefore, tissue samples should be transferred to a cold environment as quickly as possible.

II. Choose the right storage method

Although cryopreservation can maintain the integrity of RNA effectively, it may cause mechanical damage to the tissue during freezing and thawing. To mitigate this effect, protective agents such as dimethyl sulfoxide (DMSO) and glycerol can be added during the storage process to reduce the formation of ice crystals.

III. Reduce freeze-thaw cycles

Repeated freezing and thawing of tissues can lead to increased RNA degradation. Therefore, try to minimize the number of freeze-thaw cycles of tissue samples and refrozen the remaining samples after each use.

IV. Choice of fixation method

Different fixation methods can affect the integrity of RNA in tissue samples. For example, although paraffin embedding is a commonly used fixation method, the RNA content extracted is lower and is more suitable for quantitative PCR analysis.
